Melanotan 1 is a linear alpha-MSH analog with a substitution at position 4 that increases stability relative to native alpha-MSH, and is studied primarily for MC1R interaction. Melanotan 2 is a cyclic lactam analog with a broader receptor profile spanning MC1R, MC3R, MC4R and MC5R, which is why the two are used to answer different selectivity questions rather than as substitutes for one another. PT-141 is a des-amino metabolite of the Melanotan 2 structure with a shifted profile favouring MC3R and MC4R, and is listed under its research designation.
