Kisspeptin 10mg: the decapeptide behind the name

The material in this 10 mg vial is registered as kisspeptin-10 (CAS 374675-21-5), a ten-residue peptide amide whose sequence reads YNWNSFGLRF-NH2 in one-letter code. Four of those ten positions are aromatic: a tyrosine at the N-terminus, a tryptophan at position 3 and phenylalanines at positions 6 and 10. Two asparagines, at positions 2 and 4, flank the tryptophan, and all ten residues are standard L-amino acids.

Next to the polar tripeptides and acidic chains elsewhere in the catalog, this is a compact and fairly hydrophobic molecule. It is the shortest of the KISS1-derived peptides and the one most often used as a reference form, but the family name alone does not specify it. Gonadorelin, another short C-terminally amidated peptide in the catalog, is identified the same way: by its terminal amide and exact residue count.

Purity is still integrated on the trace at around 214 nm, where every peptide-bonded impurity is seen, and the 280 nm channel serves as a cross-check that under-reports any impurity lacking aromatic residues. The aromatic side chains and the leucine give solid retention on C18, so the main peak elutes mid-gradient, well clear of the solvent front.

The tryptophan indole is the most oxidation-prone site in the sequence, giving +16 and +32 Da products; the two asparagines are candidates for deamidation at +1 Da; and single-residue deletions, truncated chains and residual protecting groups are the usual synthesis-related species, several of them eluting close to the main peak. At about 1302.4 g/mol, 10 mg of pure free base corresponds to roughly 7.7 µmol. The real figure is lower, because the counter-ion and residual water in a lyophilized salt add mass without adding peptide.
