What CJC-1295 DAC is

CJC-1295 DAC is a 30-residue conjugate built on modified GRF(1-29). Its first 29 positions are the tetrasubstituted GHRH fragment (D-Ala2, Gln8, Ala15, Leu27, C-terminal amide), and a lysine is appended at position 30 whose side-chain amine is acylated with a 3-maleimidopropionyl group. That appended unit is what “DAC”, short for Drug Affinity Complex, refers to: a maleimide ring engineered as a thiol-reactive handle.

The DAC label is not decorative. It changes the molecular mass by roughly 280 Da, the retention time and the chemistry of the product, because a maleimide is an electrophile that the unmodified chain does not have. CJC-1295 without DAC, listed separately in our catalog, is a different material with its own analysis, and a record that says only “CJC-1295” cannot tell the two apart.

How CJC-1295 DAC is analyzed

The maleimide defines the impurity profile. In water the five-membered ring can open by hydrolysis, adding 18 Da and producing a maleamic acid species that usually elutes near the parent; any free thiol present can add across the double bond to give an adduct of higher mass. Those two species, together with the epimer that would arise at D-Ala2, are the peaks a method for this compound needs to resolve. The deconvoluted electrospray mass should sit near 3647, well clear of the roughly 3368 expected for the no-DAC chain.

Because the reactive group can change over time in solution, the age and conditions of any analysis are fair questions for this product in particular.
