How GLP-1 10mg is analyzed

Lipidated peptides behave differently from ordinary sequences on a column. The long diacid chain promotes self-association, so peak tailing, carryover from one run to the next and concentration-dependent peak shape are all more common than with an unmodified 31-mer. Butyl or octyl phases with 300 Å pores, or a C18 run warm, are the usual ways to keep the main peak narrow. At this size, average and monoisotopic masses differ by a couple of daltons, so a mass report should say which it quotes; the 4+ and 5+ ions fall close to m/z 1029 and 824.

Typical related substances include the peptide lacking its lipid side chain, which elutes much earlier, oxidized tryptophan, deamidated glutamine and a D-histidine epimer at the N-terminal residue, which can sit just beside the main peak.
