What BPC-157 is
BPC-157 is a linear synthetic pentadecapeptide with the sequence Gly-Glu-Pro-Pro-Pro-Gly-Lys-Pro-Ala-Asp-Asp-Ala-Gly-Leu-Val, or GEPPPGKPADDAGLV in one-letter code. All fifteen residues are ordinary L-amino acids, the chain contains no cysteine and so no disulfide, and neither terminus is capped. The letters come from a phrase in the early literature, and the number is a laboratory designation with no structural meaning.
Two features set it apart from its catalog neighbors. Four of its fifteen positions are proline, three of them in an unbroken run at positions 3 to 5, and its only acidic cluster is an Asp-Asp pair at positions 10 and 11. How BPC-157 is analyzed
The proline run is what makes BPC-157 unusual on a reverse-phase column. Each Xaa-Pro bond can adopt either a cis or a trans geometry, and the two forms interconvert over seconds to minutes, roughly the same timescale as the separation itself. The main band is therefore often wide, sometimes shouldered, even in clean material; a hotter column speeds the exchange and narrows it, whereas a genuine impurity keeps its own peak. The sequence has no aromatic side chains, so detection relies on the amide backbone near 214 nm, and electrospray MS should show a singly protonated ion close to m/z 1420 with a doubly charged ion near 710.
The Asp-Asp pair is the main chemical liability. Aspartate can cyclize to a succinimide, losing 18 Da, and reopen as isoaspartate, which has the same mass as the parent and tends to hide inside the already broad main band. Is a broad BPC-157 peak a sign of low purity?
Not on its own. Slow cis-trans exchange at the three consecutive prolines widens the band in clean material, so an orthogonal check, such as a hotter column or an LC-MS scan across the peak, is what separates conformers from a real contaminant.
Why can’t a mass spectrum alone rule out isoaspartate in BPC-157?
Isoaspartate is an isomer of aspartate with an identical mass, so the intact reading stays near 1419.5 either way. It has to be resolved chromatographically or identified with more specialized techniques.
