How GLP-1/GIP is analyzed
Two structural features pull the chromatography in different directions. The C20 lipid chain drives strong retention and late elution on reverse-phase columns, while the run of three consecutive prolines near the C-terminus can give slow cis/trans interconversion, which sometimes shows up as a broadened or shouldered main peak rather than a genuine impurity. Wide-pore phases and elevated temperature help with both. Two tyrosines, two phenylalanines and a tryptophan give clear 280 nm absorbance, but purity is still integrated on the peptide bond near 214 nm. In electrospray, look for the 3+, 4+ and 5+ ions near m/z 1605.5, 1204.4 and 963.7.
Relevant impurity classes are des-lipid peptide, tryptophan oxidation, deamidation at either glutamine, loss of the C-terminal amide (a +1 Da free-acid variant) and deletion sequences within the Gly-Pro-Ser tail.
