What Cortagen is
Cortagen is the designation used for Ala-Glu-Asp-Pro (AEDP), a tetrapeptide of four ordinary L-amino acids with an average free-acid mass close to 430 g/mol. Its identity turns on the final residue. Proline is the only standard amino acid whose side chain closes back onto its own backbone nitrogen, and placing it at the C-terminus gives AEDP a conformational and chemical character none of its close relatives share.
Those relatives are very close indeed. Remove the proline and what remains is Cartalax (AED). Swap it for glycine and the result is Epithalon (AEDG), about 40 Da lighter; swap it for arginine and it becomes Cardiogen (AEDR). Four products, one opening triplet, and only the last position and the mass to separate them.

The Asp-Pro bond drives two things an analyst should expect. First, the amide in front of proline can sit in cis or trans form, and the two interconvert slowly enough on the chromatographic timescale to broaden the Cortagen peak or leave a shoulder; that is one molecule in two shapes, and a constant mass across the peak confirms it. Second, aspartyl-prolyl bonds are comparatively labile under strongly acidic conditions, so a small signal at the Cartalax mass, near 333, is a plausible hydrolysis product worth checking for. The pyrrolidine ring gives AEDP slightly more retention than Epithalon on C18, though it is still an early eluter read at about 214 nm, with the protonated parent expected just above 430.

Usually not. Cis–trans isomerism about the Asp-Pro bond can split or broaden the peak of a single species. If the mass holds steady across the whole peak, the effect is conformational.

That mass matches Ala-Glu-Asp, the fragment left behind if the Asp-Pro bond hydrolyzes. A trace amount points to acid-driven cleavage; a large one suggests the material is not AEDP.
