What Melanotan 1 is

Melanotan 1, also written MT-1 or MT-I, is a linear synthetic tridecapeptide: Ac-Ser-Tyr-Ser-Nle-Glu-His-D-Phe-Arg-Trp-Gly-Lys-Pro-Val-NH2. It follows the thirteen-residue alpha-MSH sequence with exactly two substitutions, norleucine at position 4 in place of methionine and D-phenylalanine at position 7 in place of the L form, which is why the literature also calls it [Nle4, D-Phe7]-alpha-MSH or NDP-MSH. Its International Nonproprietary Name is afamelanotide. Both termini are capped, with an N-terminal acetyl group and a C-terminal amide.

The shared name with Melanotan 2 is misleading. Melanotan 2 is a seven-residue lactam-bridged ring of about 1024 g/mol, while MT-1 is an open chain roughly 620 Da heavier. The numeral is not a version number, and a test result for one says nothing about the other. PT-141 belongs to the same structural family but is likewise a separate cyclic compound.

How Melanotan 1 is analyzed

With a blend of hydrophobic residues (Tyr, D-Phe, Trp, Nle, Pro, Val) and three positively charged side chains under acidic conditions, MT-1 retains well on C18 and elutes mid-gradient; the ion-pairing additive has a visible effect on its peak shape. Tyrosine and tryptophan make it easy to see at 280 nm, but purity is integrated near 214 nm so that non-aromatic deletion and truncation products from a thirteen-step synthesis are counted. Electrospray spectra typically show multiply charged ions that deconvolute to a neutral mass near 1647. Because norleucine replaces methionine, the formula contains no sulfur and a +16 Da sulfoxide companion is not expected. The D-Phe residue is invisible to mass spectrometry, so an epimerized L-Phe impurity can only be revealed chromatographically as a closely eluting diastereomer.

Yes. MT-1, [Nle4, D-Phe7]-alpha-MSH, NDP-MSH and afamelanotide are all names for the same capped thirteen-residue sequence.

Why does MT-1 contain norleucine?

Norleucine matches the side-chain length of methionine but has a methylene group where methionine has sulfur. That removes the methionine oxidation pathway, so no +16 Da sulfoxide species is expected on the chromatogram or the mass spectrum.

Can a mass spectrum alone confirm Melanotan 1?

Not completely. The mass separates it cleanly from Melanotan 2 and PT-141, but a version with L-Phe at position 7 weighs exactly the same, so chromatographic separation is needed to address stereochemistry.
