What the terminal arginine changes
A guanidinium side chain is permanently protonated, so AEDR carries a positive charge alongside its two acidic residues at essentially any working pH. In practice that means ion-pairing additive in the mobile phase is load-bearing for retention, peak tailing from silanol interaction is common on silica-based columns and is not the same thing as impurity, and retention time shifts with small changes in mobile-phase composition. Retention time alone is never an identity confirmation for this compound; it has to be read alongside a mass.
Arginine-containing peptides also bind counter-ion readily, which widens the gap between chromatographic purity and net peptide content. Both numbers are needed and they answer different questions: purity tells you what fraction of the peptide present is the target sequence, content tells you how much peptide is in the vial at all.
Detection
AEDR has no aromatic residue, so 280 nm detection is unavailable and the analysis must be read at approximately 214 nm. A published purity figure without a stated wavelength and gradient cannot be compared against another vendor’s figure.

Column chemistry and dimensions, mobile phase and gradient including the ion-pairing additive, detection wavelength, retention time, integrated peak area with a visible integration window, and mass confirmation consistent with a 489 g/mol free acid. Then, separately, net peptide content and the counter-ion form.
