Ion pairing carries the retention. Without an ion-pairing additive in the mobile phase, a small peptide with a permanent positive charge and two acidic residues has almost nothing to hold it on a C18 column. Trifluoroacetic acid is usually doing the work, which means the method description is not a formality here — it is the reason the peak is where it is.

Silanol interaction produces tailing. A basic side chain on a silica-based column interacts with residual free silanols. Tailing on a Pinealon peak is frequently a column artefact rather than evidence of an impurity, and reading it as impurity leads to the wrong conclusion about the material.

Retention is pH-sensitive. Small changes in mobile-phase composition move the peak. Two suppliers can therefore publish different retention times for identical material, which is why retention time on its own is never an identity confirmation and has to be paired with a mass.

Detection

EDR contains no tryptophan, tyrosine or phenylalanine, so there is nothing to see at 280 nm. The analysis has to be read at approximately 214 nm on the peptide bond, where the mobile phase absorbs more strongly and baseline quality matters. A purity figure quoted without a wavelength is not evaluable for this compound.

Names in this family are designations, not identifiers
