What Epithalon is

Epithalon, also spelled epitalon, is the catalog name for the synthetic tetrapeptide Ala-Glu-Asp-Gly, abbreviated AEDG. All four residues are standard L-amino acids, the chain ends in a free carboxylic acid, and the free-acid form has the formula C14H22N4O9 with an average mass near 390 g/mol. Of all the coined designations in our short-peptide group it is the one searched for most often, which also makes it the one most often mislabeled elsewhere.

Epithalon should not be confused with epithalamin, a term for a peptide preparation rather than one defined sequence. Inside our own catalog its closest relatives share its opening three residues: Cortagen replaces the terminal glycine with proline, and Cardiogen replaces it with arginine. Livagen uses the same amino acids in a different order behind a lysine. The printed sequence, not the word on the label, is what should be compared.

How Epithalon is analyzed

The feature worth knowing in AEDG is the Asp-Gly junction at positions three and four. Aspartate followed by glycine is the motif most prone to cyclizing into a succinimide, which can reopen as isoaspartate: a species with exactly the parent’s elemental composition, so mass spectrometry cannot pick it out by m/z alone. On a reverse-phase trace it tends to run as a small peak hugging the main one, which is why a shallow gradient with a low starting organic fraction matters for this sequence. The succinimide intermediate, when present, reads about 18 Da light. With no aromatic side chain, the peptide is quantified on the amide band in the low-UV region, and a protonated ion just above 390 is the expected identity signal.

Epithalon questions

Are epitalon and epithalon different compounds?

No. They are two transliterations of one name and both refer to AEDG. The sequence and the 390 g/mol mass are the same whichever spelling a supplier prints.

Can a mass spectrum rule out isoaspartate in Epithalon?

Not on its own. Isoaspartyl AEDG has the same composition as the native peptide, so it has to be separated chromatographically, where it usually elutes as a close neighbor of the main peak.

How is Epithalon different from epithalamin?

Epithalon is a single synthetic tetrapeptide with a fixed sequence. Epithalamin names a peptide preparation, so the two terms are not interchangeable.
